VELANDIA,Sindi Alejandra; QUINTERO,Elizabeth; STASHENKO,Elena E; OCAZIONEZ,Raquel E.

Bioisosteric modification on melatonin: synthesis of new naphthalene derivatives, in vitro antioxidant activity and cytotoxicity studies BJPS
Shirinzadeh,Hanif; Ghalia,Mohammed; Tascioglu,Alev; Adjali,Ferial Intissar; Gunesacar,Gulsen; Gurer-Orhan,Hande; Suzen,Sibel.
Melatonin (MLT) is a strong free radical scavenger that protects the body from the deleterious effects of excess oxidants. Synthesis of MLT analogue compounds with antioxidant potency has recently attracted the interest of researchers. In general, the strategy consists of modifying the groups in the different sites of the indole ring or replacing the indole ring with an analogue. As part of our ongoing research, the antioxidant capacity and cytotoxicity of newly synthesized MLT analogue naphthalene derivatives were evaluated. The radical scavenging activity was tested by a 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Most of the synthesized compounds showed significant antioxidant activity in comparison to MLT. The structure-activity relationship was...

Cytotoxic evaluation of essential oil from Casearia sylvestris Sw on human cancer cells and erythrocytes Acta Amazonica

Effects of plant lectins on in vitro fibroblast proliferation BABT
Sell,Ana Maria; Costa,Celso Paulino da.
Lectins are carbohydrate-binding proteins that have been isolated from various sources and presented a wide spectrum of biological activities. The effects of four lectins, namely, Phaseolus vulgaris phytohemagglutinin, PHA, wheat germ agglutinin, WGA, Artocarpus integrifolia seed lectins, jacalin and artocarpin, on in vitro fibroblasts proliferation were investigated. The lectins did not influence the initial cell adhesion to the plate. PHA and WGA at 10-20 µg/mL concentrations significantly decreased fibroblasts proliferation. At these concentrations, they caused morphological alterations on cells and over 80 µg/mL, promoted cell death. Neither jacalin nor artocarpin significantly affected cell proliferation.

Human mesenchymal stem cells are resistant to cytotoxic and genotoxic effects of cisplatin in vitro Genet. Mol. Biol.
Bellagamba,Bruno Corrêa; Abreu,Bianca Regina Ribas de; Grivicich,Ivana; Markarian,Carolina Franke; Chem,Eduardo; Camassola,Melissa; Nardi,Nance Beyer; Dihl,Rafael Rodrigues.
Abstract Mesenchymal stem cells (MSCs) are known for their important properties involving multilineage differentiation potential., trophic factor secretion and localization along various organs and tissues. On the dark side, MSCs play a distinguished role in tumor microenvironments by differentiating into tumor-associated fibroblasts or supporting tumor growth via distinct mechanisms. Cisplatin (CIS) is a drug widely applied in the treatment of a large number of cancers and is known for its cytotoxic and genotoxic effects, both in vitro and in vivo. Here we assessed the effects of CIS on MSCs and the ovarian cancer cell line OVCAR-3, by MTT and comet assays. Our results demonstrated the resistance of MSCs to cell death and DNA damage induction by CIS,...

Viability in protoplasts and cell suspensions of Coffea arabica cv. Catimor Electron. J. Biotechnol.
Fernandez-Da Silva,Rafael; Menéndez-Yuffá,Andrea.
Two methods were evaluated in order to assess the viability of protoplasts and cell suspensions of Coffea arabica cv. Catimor used in a protocol of transformation by electroporation. One method consisted of staining with 1 % Evans blue and the other staining with 1% 3-[4,5-dimethyltiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT). When Evans blue was applied to viable cells and protoplasts they either did not stain or acquired a faint blue colour. However, it was difficult to distinguish the non-viable cells in the wide spectrum of clear blue tonalities. In contrast, with the MTT assay only the viable cells and protoplasts reduced this salt to the red coloured formazan; viable and non-viable cells were distinguished more clearly with MTT than with Evans...
